SRX22404042: Symphalangus syndactylus raw sequence reads

Design: The UHMW DNA was extracted from frozen cell pellets using the Circulomics Nanobind UHMW DNA Extraction Protocol and assessed for fragment size using a pulsed field gel. The fragments span from 50 Kb to 1000 Kb. Genomic DNA libraries were prepared using the "(ONT/CirculomicsLibraryProtocol)SQK-ULK001-ULK_9124_v110_revA_24Mar2021-promethion.pdf protocol from Oxford Nanopore. Each library was loaded onto a FLO-PRO002 flow cell and run for 72 hours with two subsequent loadings at 24-hour intervals. The libraries were sequenced on a PromethION (Oxford Nanopore) running MinKNOW software version 21.05.13. Basecalling and modified base detection (5mC) were performed using Guppy 6.3.8.
